Anti-Fade Mounting Medium (With PI) is a glycerol-based mounting medium containing an anti-fluorescence quenching reagent. It can be used to seal tissue or cell slides for fluorescence-related applications. After mounting, slides can be observed for several weeks without drying out or requiring sealing. This medium can help prolong fluorescence and maximise fluoresence intensity, and assist with antigen-antibody binding. This medium is compatible with a wide range of fluorophores and fluorescent proteins, such as blue fluorescent protein, FITC, Cy2, Cy3, Cy5, Cy7, DAPI, rhodamine, Texas Red, AMCA, R6G, Hoechst 33258, Alexa Fluor (AF), and more. This medium contains Propidium Iodide (PI), an intercalating agent that can be used to stain cells. It can be used as a DNA stain for flow cytometry, evaluating cell viability, determining DNA content in different stages in the cell cycle, differentiating between necrotic, apoptic and normal cells, and visualising the nucleus and other DNA-containing organelles. When PI is bound to nucleic acids, the fluorescence excitation maxiumum is 535 nm and the emission maximum is 617 nm.
